中文摘要: 基于非任务态功能磁共振成像的人脑功能连接方法已经被广泛用于精神分裂症研究。多数研究基于数据采集时间内功能连接恒定不变这一假设,因此,只能提供一个“静态”的连接模式。然而,人脑是不断活动的,其功能连接也是不断变化的,研究功能连接的动态变化可以更加准确地刻画脑疾病患者功能连接的异常改变,有助于发现敏感性高、特异性强的影像学标志。为此,本项目首先采用多尺度动态功能连接方法(动态短程功能连接、动态长程功能连接以及动态全脑功能连接网络)全面研究精神分裂症功能连接的异常变化模式,寻找患者变化最显著的动态功能连接指标。在此基础上,利用模式识别技术,测试基于单个或多个动态功能连接指标诊断精神分裂症的准确性。通过上述研究不仅可以发现新的精神分裂症特异性的生物学标记,还可以建立基于动态功能连接的精神分裂症个体化诊断模型。该项目的实施有助于提高对精神分裂症脑功能连接异常的认识,为该疾病的诊断提供方法。

英文摘要: The non-task functional magnetic resonance imaging (fMRI) based functional connectivity method has been widely used in schizophrenia. Most of these studies are based on the assumption that functional connectivity is constant during the observation period, thus providing a static pattern of brain activity coherences. However, given the time dependent and dynamic nature of brain activity and functional connectivity, investigation of the dynamic change can depict the alteration of functional connectivity more accurately in patients with brain disease and further help us to find the neuroimaging marker with high sensitivity and specificity. To this end, this project aims to adopt multiscale dynamic functional connectivity method (dynamic short-range functional connectivity, dynamic long-range functional connectivity and dynamic whole-brain functional connectivity network) to comprehensively investigate the abnormal changed pattern of functional connectivity in schizophrenia and seek for the most changed index of dynamic functional connectivity. Based on these, pattern recognition technique is used to test diagnostic accuracy of single or multiple dynamic functional connectivity index. In this way, we can not only find novel specific biomarkers for schizophrenia but also construct a diagnostic model for schizophrenia at the individual level. Overall, this project may improve our understanding of abnormal functional connectivity in schizophrenia and provide method for disease diagnosis.
