中文摘要: 跨膜多肽和蛋白在生物膜中的正确取向是其具有生物活性的先决条件,但是目前相关的系统性研究还非常缺乏,找出决定跨膜多肽和蛋白取向的主要因素,并揭示其作用的规律和机理,具有重要的科学意义。本项目首先拟利用具有长荧光寿命的荧光分子DBO作为荧光探针,以色氨酸作为淬灭剂,利用DBO荧光寿命和荧光强度的变化,建立一个简单有效的检测单螺旋跨膜多肽在模拟生物膜中取向及其分布的方法;然后利用这一方法对不同跨膜多肽的取向进行表征和比较,系统地探讨影响单螺旋跨膜多肽在膜中取向的各种因素,如氨基酸序列、肽链长度、氨基酸带电性、生物膜构成以及生物膜曲率等;最后,在这些工作的基础上提出单螺旋跨膜多肽在生物膜中采取正确取向的机理模型。这一工作将为深入认识跨膜多肽和蛋白的折叠过程提供重要的实验和理论依据,为进一步揭示膜蛋白的折叠机理奠定基础。

英文摘要: Correct orientation of transmembrane peptides and proteins in membrane is essential for them to obtain biological functions. However, the mechanism determing how they orientate is not yet clear. It is therefore very important to reveal the key factors controlling the orientation of transmembrane peptides and proteins in membrane, and to investigate their woking mechanism. For this purpose, DBO, a small hydrophilic fluorescent molecule with very long fluorescnece lifetime, will be applied as fluorescent probe and tryptophan as quencher to establish a novel method to characterize the orientation of single-helix transmembrane peptides in membrane. With this method, factors influencing the orientation, such as the sequence, length and charge of peptides, as well as the composition and curvature of the mimic membrane, will be systematically investigated. The mechanism determining the orientation of transmembrane peptides in membrane will be proposed based on the experimental data. This work will provide important information experimentally and theoretically for further understanding the folding mechanism of membrane proteins.
