中文摘要: 反相高效液相色谱是分离分析核酸包括DNA、RNA和寡核苷酸最合适也是最重要的手段之一。本项目研究在不同的流动相体系中,核酸的反相高效液相色谱保留行为和结构信息之间的内在联系。通过大量的实验,系统地探索不同碱基数量、碱基组成、碱基顺序和相对位置、分子形状和二级结构等因素对保留值的贡献,归纳这些因素对不同流动相溶液中核酸存在状态和总体疏水性的影响,探讨核酸分子与固定相之间的作用机理,建立相应的数学模型,从而更加准确地描述核酸的保留行为与各级结构之间的关系。本研究结果不但可为快速选择核酸的最佳色谱分离分析方案提供可行的理论指导,而且可为实验研究核酸在溶液中的存在状态和结构行为提供一种新颖的策略。

英文摘要: Reversed-phase high performance liquid chromatography is one of the most suitable key technologies for the separation and analysis of nucleic acids including DNA, RNA and oligonucleotides. In this proposal, the intrinsic relationship between retention behavior and structure of nucleic acids will be investigated in different mobile phase systems. Through abundant experiment, we will systematically explore the contribution of base number, base composition, base sequence, relative position, molecule shape and secondary structure of nucleic acids on their retention. The influence of these factors on existing state and overall hydrophobicity of nucleic acids will be summarized, the interaction mechanism of nucleic acids with the stationary phase will be elucidated, and the related mathematical model will be established to describe the relationship between retention behaviors and structures of nucleic acids with accuracy. The reversed-phase liquid chromatographic behavior study of nucleic acids can not only provide a feasible theoretical guidance for the rapid selection of optimum chromatographic separation and analysis scheme, but also propose a novel strategy for the study of existing state and structure of nucleic acids in various solutions.
