中文摘要: 在胚胎发育、组织损伤修复等过程中,内生性电场对细胞运动等行为有广泛影响,细胞膜受体和胞内信号通路以及钙离子均对细胞运动有调控作用。本课题组研究发现电场能诱导细胞内钙离子浓度升高和极化分布,钙离子螯合剂可以抑制电场中细胞运动和形状改变,而膜受体和信号通路的抑制剂只能抑制细胞运动,不能抑制细胞形变。所以我们推测钙内流是电场诱导细胞运动的信号起点。另外还将业内公认的平面作用装置发展成毛细管曲面,更接近机体环境。为了验证钙离子的启动作用,本项目应用上述两种实验装置,结合钙离子和细胞膜受体、细胞内信号通路,采用荧光探针和免疫组化等方法标记胞内与运动相关信号分子,可视化跟踪观察电场作用后:① 钙离子与EGFRs、PI3K、PTEN、MAKP、Rho GTPases和细胞运动的关系。② 钙离子、钙离子通道和细胞运动的关系。③ 钙离子、细胞骨架和细胞运动的关系。为阐明电磁场的治病机理和临床应用奠定基础。

英文摘要: Researches have been found that during fetation and pathology course, such as tissue damage restoring, electric fields play an important role in cell biological behavior. Cell membrane receptors and intracellular signaling pathways and calcium ion are involved. We found that electric fields induce calcium ion colocalize and redistribute asymmetrically. EDTA chelator of calcium ion could inhibit cell migration and morphological change, while inhibition of membrane receptors and intracellular signaling pathways could only abrogate the migration but not the morphological change. So we presumed that the initial point of movement result from the signal of calcium ion. Moreover we improved planar direct current interference device to curved surface which simulated invivo better.To test this assumption, in this project calcium ion, membrane receptors and intracellular signaling pathways will be labelled by fluorescent probe, Immunohistochemistry and fluorescent protein to investigate: 1. the relations of calcium ion, EGFRs, PI3K/PTEN,MAKP and Rho GTPases.2. the relations of calcium ion, calcium channel and cell movement. 3. the relations of calcium ion, cytoskeleton and cell movement.
