中文摘要: 本项目在前期研究中发现的纳米颗粒集体组装结构可调控细胞生长形态和功能的基础上,针对不同尺度上纳米组装结构的有序性和能量变换性质对细胞功能的影响这一核心问题,提出在基于模板的刻蚀产生的不同尺度组装区域上利用磁场组装非磁性的金纳米壳颗粒形成有序结构的设想。研究在磁场作用下金纳米壳颗粒形成有序组装结构的过程以及磁力和组装区域尺度对组装结构有序性的协同调控作用,通过金纳米壳组装体的集体光学性质研究不同尺度下组装结构中单体间的耦合作用并进而研究组装结构调控的集体光热转换性质,最后基于金纳米壳颗粒的组装结构和光热转换性质研究不同尺度上的金纳米壳组装结构对NK细胞杀伤肿瘤的功能增强作用,探讨这种增强作用的物理化学机制。本项目的开展将有利于深化磁场控制非磁性纳米颗粒形成有序组装结构的研究,扩展大规模有序纳米结构在生物医学领域中的应用,为功能纳米结构介导的电磁场对细胞功能的调控研究打下物理化学基础。

英文摘要: In the last grant, the aggregates of nanoparticles were discovered to be capable of regulating the growth and the function of osteoblastic cells and NK cells. The key issue here is effect of the morphology as well as the transduction efficiency on the interface between the aggregates and the cells. Therefore, we proposed to combine the top-down and the bottom up approaches to assemble the gold nanoshells in the multi-spatial scale to get the order assemblies. In this project, we are to study the formation of the multi-spatial scaled areas for assembly by slectively etching the hydrophobic self assembled monolayers, the process of field mediated assembly of gold nanoshells in the different spatial scales by the light scattering, and the modulation of spatial scale on the assembled morphology. Then we will investigate the plasmonic coupling of individual nanoshells by the optical characterization and the corresponding photo-thermal property. Finally, we will explore the effect and the physichemical mechanism of the colloidal assemblies on the NK cells based on the above-mentioned results. Our project will be valuable in the biomedical application of gold nanoshells and promote the attentions to the role of assemblies morphology in the research of interaction between nanoparticles and cells.
