中文摘要: 肿瘤干细胞miRNA是肿瘤干细胞增殖、分化及凋亡的重要调控因子,它的异常表达与肿瘤的发生、发展、转移、耐药及复发等过程密切相关。肿瘤干细胞miRNA正成为肿瘤诊断和治疗的新靶点。本项目针对肿瘤干细胞miRNA的检测及治疗开展研究,结合分子探针技术、生物技术以及化学、酶学、肿瘤学等多学科知识,利用聚合引物识别、脱氧核酶及核酸回文结构技术三大平台,设计研制具有检测和酶切双功能的新型自组装杠铃荧光探针,构建基于新型双功能杠铃探针的等温链置换循环放大反应平台,建立高灵敏、高选择、方便快捷的miRNA检测新方法,开展肿瘤的酶切治疗研究及肿瘤干细胞miRNA 调控机理研究,为肿瘤预防、早期诊治、预后判断及治疗提供临床指导作用。

英文摘要: MicroRNA( miRNA) is a kind of short, noncoding RNAs that powerfully regulate gene expression at the post transcriptional level. It was revealed that miRNA regulated cell proliferation, differentiation, apoptosis and various physiological and pathological pathways in tumor stem cell. MicroRNA's abnormal expression in tumor stem cell is closely related to the process of tumor occurrence, development, metastasis, drug resistance, recurrence, etc. MiRNA of tumor stem cell is a new target for diagnosis and therapeutic tumor. Detection and expression analysis of miRNA are significant to the understanding of functional mechanism of miRNAs and the diagnosis. Aimed to detection and treatment research of the tumor stem cell miRNA, this project mainly includes three aspects: Design and construction of a detection and cleavage dual-functional self-assembly barbell fluorescent probe based on three platforms, such as polymerization primer recognition, DNAzymes and DNA palindrome structure technology; establishing fluorescence signal amplifying detection system of isothermal circular strand-displacement polymerization reaction based on the novel dual-functional barbell probe and development of new methods with high sensitivity, high selection, high convenience and efficiency for the detection of miRNA; research of miRNA regul
