中文摘要: 口蹄疫(FMD)是由FMD病毒(FMDV)引起的一种严重危害猪、牛等偶蹄动物养殖业的烈性传染病。由于FMDV的高度变异性、众多血清型以及持续性感染,致使本病通过疫苗免疫手段难以有效防控。通过调控病毒诱导细胞凋亡手段,可防控病毒病的扩散和传播。本项目拟在已初步证明FMDV VP2基因可诱导宿主细胞发生凋亡,且VP2可与凋亡基因Siva结合的基础上;通过高表达和RNAi沉默技术,上调和下调Siva基因的表达水平,检测其在VP2诱导细胞凋亡过程中的作用,探究VP2诱导凋亡的信号通路及分子机制;突变VP2的不同位点,利用pull-down等技术,精确定位VP2诱导凋亡的功能位点;应用反向遗传技术,在病毒水平验证诱导凋亡的功能位点变异后VP2对细胞凋亡的影响。本项研究将为研发特异性抗FMDV药物提供靶标,并为获得无细胞凋亡作用的高效价疫苗毒株奠定基础。

英文摘要: Foot-and-mouth disease (FMD) is a highly contagious and economically devastating disease of cattle, swine and other cloven-hoofed ruminants. It is difficult to control because of variability and several serotypes and persistent infection. The study will be developmented on the base of the VP2 inducing host cell apoptosis of FMDV and binding to apoptosis protein siva. Siva will be up-regulation and down-regulation via overexpression or RNA interference technique, and the role of siva in the VP2 inducing apoptosis will be determined. Mutation at different sites of the VP2 , will be made pull-down and other technique can be used for precisely evaluating the sites of the VP2 which induced apoptosis; and then affection of apoptosis will be analyzed when functional sites of VP2 were mutated by reverse genetic technique in virus levels. This study will provide the potential target for anti-FMDV drug, and obtain virus strain which don't lead to apoptosis of the cultured cells, thereby increase vaccine potency.
