中文摘要: 磁共振成像(MRI)已成为当代临床诊断中最有力的检测手段之一。传统造影剂主要包括顺磁小分子配合物和超顺磁性纳米氧化铁等两类化合物,它们虽然得到较快发展,但在在灵敏度、安全性、成本、剂量、靶向性等多个方面仍有很大的改进空间,开发新型MRI造影剂依然是该领域的热点。本项目使用亲水基团修饰过的多齿含氧配体和铁盐反应得到一系列具有良好水溶性的多核铁(III)氧簇化合物,拟将这些结构确定的铁(III)氧簇合物用于MRI造影剂的研究和开发。这种新型造影剂磁性中心的尺寸介于单个顺磁性离子和纳米粒子之间,有望结合传统顺磁性和超顺磁性MRI造影剂的优点,从而具有高度可控的结构和尺寸、相对简单的制备过程、优良的成像效果和低毒性等特点。该课题的研究成功,不仅可以大大扩展多核铁(III)氧簇化合物的应用范围,更是为新型MRI造影剂的设计合成提供了新的思路。

英文摘要: Magnetic Resonance Imaging (MRI) has become one of the most powerful technology in modern clinical diagnosis. Traditional agents mainly include paramagnetic samll-molecule coordination complex and super paramagnetic iron oxide nanoparticle. Although these agents have been highly develped, there still need many improvements in many aspects including sensitivity, safety, cost, dosage and target of the existing MRI constrast agents, and developing new agents is still attracting. This project aims at synthsis of a series of hydrosoluble multiple nuclear iron(III)-oxo cluster complexes from the decorated multidentate oxygen-containing ligands and iron salt, and the obtained cluster complexes can be used as MRI constrast agents. The size of magnetic core in these agents falls within the range of single paramegnetic ion and super paramegnetic nanoparticle, and they may possess the advantages of both conventional paramagnetic and super paramagnetic MRI constrast agents, which include highly controllable structures and sizes, relative simple syntheses, good imaging and low toxicity etc. The success of this project will not only extend the application of the iron-oxo clusters, but also supply new sight of designing and synthesizing novel MRI constrast agents.
