中文摘要: 髓系来源的抑制性细胞(MDSCs)是一群被认为未完全分化成熟的髓系细胞。在肿瘤等病理条件下,MDSCs发生扩增和活化,表现为抑制免疫、促进肿瘤细胞生长和转移以及血管生成等。申请人通过骨髓移植小鼠肿瘤模型发现了骨髓细胞中SENP1缺失能够导致肺和肝中MDSCs显著增多,并促进肿瘤细胞发生肺和肝转移。因此,我们提出肿瘤发生时髓系细胞中SENP1表达下降促进MDSCs的扩增与活化,从而促进肿瘤的转移。本课题将在前期研究的基础上,主要研究如下问题: 1. SENP1如何调控MDSCs的扩增与活化?2. SENP1缺失的MDSCs有什么样的功能特征?3. 在肿瘤病人标本中,MDSCs中SENP1的表达与结肠癌转移等临床指标是否有关联?这些研究将使我们能更深入地了解SENP1调控MDSCs的作用及机制,为靶向MDSCs的肿瘤治疗策略提供理论依据。

英文摘要: Myeloid-derived suppressor cells (MDSCs) represent a heterogeneous population of activated immature myeloid cells that consists of myeloid progenitors and precursors of macrophages, granulocytes and dendritic cells. MDSCs have the ability to support tumor progression by promoting tumor cell survival, angiogenesis and metastasis. In previous studies, we found that the deficiency of SUMO-specific protease 1 (SENP1) in bone marrow cells promotes the accumulation of MDSCs in lung and liver, and promotes metastasis in these two organs in a tumor metastasis mouse model. Therefore we propose that the decreased SENP1 in tumor-bearing hosts promote MDSCs expansion and activation, and then promotes tumor metastasis. In this study, we will focus on these three questions: 1. What is the mechanisms that SENP1 regulate the expansion and activation of MDSCs? 2. What is the functional characteristic of SENP1 deficient MDSCs? 3. What is the relationship between SENP1 expression in MDSCs and tumor progression in colon cancer patients? This project will make us further understanding the role of SENP1 on MDSCs and will contribute to the novel opportunities for targeting MDSCs.
