中文摘要: 苯中毒是我国重点防治的职业病,主要引起血液毒性。本课题组前期在慢性苯中毒患者中发现了与苯血液毒性密切相关的、新的长链非编码RNA(lncRNA)-VNN3和OBFC2A,但其功能和作用机制有待阐明。本研究拟通过体外实验和人群研究相结合,采用RNA pull-down、RIP-chip、ChIRP-seq方法,研究与VNN3和OBFC2A相互作用的蛋白和DNA;采用esiRNA干扰与FISH 定位相结合的c-KLAN技术抑制lncRNA功能后,研究上述lncRNA定位情况及其对苯毒性相关基因甲基化(PARP-1、PTEN、STAT3)及microRNA(miR-205、miR-133a)异常表达的调控作用及其机制,旨在从调控基因表达和表观遗传修饰两方面阐明lncRNA调控苯毒作用的机制;进一步在苯中毒、苯接触人群中验证血浆lncRNA的表达变化情况,为苯中毒危害筛查和早期诊断提供新靶标。

英文摘要: Benzene poisoning, which mainly causes hematotoxicity, is the priority of the prevention and control of occupational diseases in china. Rencently, we screened out two key lncRNAs (VNN3&OBFC2A) associated with benzene poisoning through high-throughput lncRNA microarray. However, the function and mechanism of lncRNAs in benzene hematotoxicity remain poorly understood. Constructed overexpression vector and endoribonuclease-prepared siRNA(esiRNA)of these lncRNAs, combined knockdown and localization analysis of noncoding RNAs(c-KLAN), RNA-pull down, RNA-binding protein immunoprecipitation(RIP) and chromatin isolation by RNA purification-sequence (ChIRP-seq) will be applied to explore function of these lncRNAs in benzene hematotoxicity and its molecular mechanisms. Furthermore, these screened lncRNAs will be validated by using qRT-PCR in people exposed to benzene and benzene poisoning patients. LncRNA provides a rationale for the potential development of lncRNA-targeted approaches for the prevention and early diagnosis of benzene poisioning.
