中文摘要: 提出一种新的光学无损的鼻咽癌早期诊断和分型的方法。光学相干层析成像技术(OCT),可以对生物组织内部的微观结构进行高分辨率的横断面层析成像。OCT的分辨率约为10um,扫描深度可达1-3mm。拉曼光谱技术(RS)可以提供丰富的分子结构特征和物质成分信息,通常被称为物质的分子指纹。拉曼光谱对肿瘤生长和发展过程中细胞分子的变化具有高灵敏度和高分辨率的特点。本项目首次结合OCT与RS技术,即形态学和分子谱结合,用于鼻咽癌早期诊断和分型的研究。对鼻咽癌的不同病变阶段及不同分型的活检组织进行OCT扫描成像和光谱采集,分析它们的结构和成分的变化,找到早期诊断和个性化特征的定性和定量的指标,并建立计算机的自动识别模型,从光活检角度实现鼻咽癌的早期诊断和个性化诊断。两种技术都是无损,实时,分辨率高的优点,且都能与光纤结合,有望实现在体、无损的临床诊断一体化工具。

英文摘要: We propose a new noninvasive method of early diagnosis and typing of nasopharyngeal carcinoma(NPC) using optical coherence tomography(OCT)and Raman spectroscopy(RS). OCT, a cross-sectional tomography modality for the microstructure of tissue, with the resolutions less than 10 μm; depth up to 1-3 mm. RS can provide the information of molecular structure and component, ofter referred to as the molecular fingerprint of the molecular. RS has high sensitivity and resolution of cellular and molecular changes of tumor growth and development. Thus, our project, combined with OCT and RS technique, with the characteristics of morphological image and molecular spectroscopy, study on the early diagnosis and typing of NPC. We obtain the OCT imagings and spectrum of different types and different pathological stages of nasopharyngeal biopsy tissue, analysis their structure and component changes, find qualitative and quantitative indicators of early diagnosis and personalized features, establish the computer automatic recognition model; implement early and personalized diagnosis of NPC in respective of "optical biopsy". The two techniques are nondestructive, real-time, sensitive and can be combined with optical fiber, which is expected to achieve in vivo, non-destructive integration of clinical diagnostic tool.
