中文摘要: 以芽殖酵母细胞信息素信号网络为研究对象,对该生物网络进行数学建模并分析其时空动力学行为,理论上研究细胞命运如何对外界刺激变化做出抉择。首先基于反应动力学以及该生物网络的相关实验数据,建立包含细胞膜、细胞质以及细胞核空间生化反应的数学模型,研究细胞质中活性激酶信号的确定性动力学行为,从而得到系统对信息素变化的响应;然后考虑支架蛋白质Ste5固定在细胞膜附近以及细胞质内各种激酶的扩散这两个关键的空间效应,建立反应扩散模型,研究活性激酶信号传播的时空动力学特点。本项目的研究意义在于:在生物问题驱动下开展复杂系统的研究,建立整个信号网络的完整模型,利用非线性动力学分析和数值模拟揭示信号转导的时空动力学性质,从而加深对细胞命运抉择的系统理解,为癌症和发育缺陷疾病的治疗提供理论依据,并希望为实验提供定量预测。

英文摘要: In this application, we investigate the budding yeast pheromone signaling network. We construct mathematical models and analysis dynamic behavior about this biological network, further show how cells enter different life cycle stage in response to environmental change theoretically. Based on reaction kinetics and experimental data about this biological network, we built a mathematical model which includes the biochemistry reactions from the plasma membrane to the nucleus via the cytoplasm, then using this model we study the deterministic dynamic behavior about active kinases in the cytoplasm, and obtain the response of the system to pheromone change. To better understand this signal transduction network, we construct a reaction-diffusion model which considers the scaffold protein Ste5 near the plasma membrane and diffusion process of active kinases in the cytoplasm, to explore spatiotemporal behavior of active kinases. The meaning of our application is as follows: we use the theory of complex system to explore the biological problem and model the whole biological network. Moreover, we study the spatiotemporal behavior of signal transduction by using nonlinear dynamic analysis and numerical simulation. This makes us to understand further about selecting cell fate.
