中文摘要: 水稻条纹病毒(Rice stripe virus, RSV)引起的水稻条纹叶枯病是我国当前水稻生产最具威胁的病害之一。前期研究发现:(1)RSV能够侵染本氏烟并呈现典型的花叶症状;(2)酵母双杂交和BiFC实验初步检测包括RSV NS3在内的多种水稻病毒蛋白能够与 植物翻译延伸因子1A(eEF1A)相互作用。本项研究利用RSV能够侵染本氏烟的特点,通过荧光共定位、UV诱导的交联耦合、CoIP或Pulldown等实验进一步验证水两种寄主植物eEF1A与RSV的相互作用,通过实时定量PCR、过表达和RNAi eEF1A的水稻或本氏烟等实验来分析寄主植物eEF1A缺失或过表达对RSV侵染、复制和增殖的影响,解析eEF1A与病毒互作的方式和意义,探讨水稻条纹病毒利用eEF1A的策略或途径。研究结果可以为深入研究RSV的基因功能和致病机理奠定基础,也可为水稻条纹叶枯病的防治提供新思路。

英文摘要: Rice stripe disease, caused by Rice stripe virus (RSV), is one of the most economically important diseases in rice plant in China. Our previous research found: (1)RSV could infect Nicotiana benthamiana and induce typical mosaic symptoms; (2) Many virus encoded proteins,including RSV NS3, could interact with eukaryotic translation elongation factor 1A(eEF1A) from Oryza sativa. In this proposal, the interactions of eEF1As from two host plants with RSV encoded proteins or genomic nucleic acids will be further validated by yeast two-hybrid system, BiFC,UV-induced cross-linking, CoIP or Pulldown tests. In addition, the influences of overexpression or RNAi of eEF1A on RSV infection and replication will be analyzed by real-time quantitative PCR technique. Based on the above research, the role of eEF1A in viral multiplication would be illuminated preliminary. The study not only could lay the foundation for further functional analysis and pathogenicity mechanism of RSV, but also could provide new ideas for the control of rice stripe disease.
