中文摘要: 低毒病毒-板栗疫病菌系统是研究丝状植物病原真菌和病毒-宿主相互作用的优秀模式系统。低毒病毒感染后,导致宿主菌丝色素减少、产孢量降低,同时发生真菌低毒力现象。已有相关研究报道表明,致病力降低的真菌菌株初级代谢产物与正常菌株相比有较大的改变,但并未明确低毒力现象与特定代谢通路之间的关系。本课题拟在已有的研究基础上,使用板栗疫病菌野生强毒株和另外3株不同毒力的低毒病毒侵染株为实验材料,通过精细分析,明确在不同毒力菌株中,受低毒病毒调控的真菌代谢物的变化规律和变化趋势,找到与之相关的代谢通路和受调控的关键基因,研究这些代谢通路在病毒侵染过程中发挥的重要作用,结合转录组和蛋白组研究数据,通过基因功能研究,找到病毒调控真菌毒力的途径和方法,明确病毒调控代谢组及其与转录组、蛋白组调控的关系,为深入研究植物病原真菌的致病机理和防控措施奠定基础。

英文摘要: The hypovirus-Cryphonectria parasitica system is an excellent model system for the study of filamentous plant pathogenic fungi and virus-host interaction. The hypovirus infection caused reduced pigment, decreased conidiation and the phenomenon of hypovirulence. The previous research showed that primary metabolites of Cryphonectria parasitica changed greatly upon hypovirus infection, but the relationship between fungal virulence and special metabolic pathways was still unclear. Based on the present studies, this research program plans to analyze the changing tendency and regular pattern of fungal metabolites using wild-type and three different hypovirus-infected strains as experimental materials to find the related metabolic pathways and key genes, confirm the important roles of these pathways during virus infection. Combined with transcriptomic, proteomic data and study on related gene functions, the experimental result will help us to better understand the hypovirulence-related metabolic pathways, learn the relationship of virus regulation between metabolomic, transcriptomic and proteomic level, lay the foundation for the deep research of fungal pathogenesis and biological control.
