中文摘要: 受能快速止血的传统中药金毛狗蕨茸毛微结构启发,本课题提出一种新型的具有可控竹节状中空微结构的壳聚糖超细纤维膜,以用于快速凝血及药物包载控释。该超细纤维膜独特的竹节状中空微结构通过微流控技术构建新型的内含液滴的管状三相层流体系为模板来构造;基于其壳聚糖成膜基材优良的促凝血功能,结合竹节状微结构对血液流动的限制作用,可协同实现快速凝血;同时,其竹节状微空腔可分隔包载药物并经缓释或快速释放直接作用于伤口,以在止血同时兼备治疗的功能。课题重点研究该超细纤维膜的微流控制备及其结构调控方法,确定其快速凝血的行为机理及其与内部微结构的关系,考察其药物包载和控释特性及其可控性、并建立相关传质动力学模型,研究药物控释与凝血功能的协同作用和机制。研究结果将为改进现有医用纤维材料的凝血功能并赋予其多功能化开辟新途径,为新一代超细中空纤维膜的设计与可控制备提供理论基础。

英文摘要: Inspired by the traditional Chinese medicine Cibotium barometz with bamboo-like microstructure for fast hemostasis, here we report on a novel type of ultrathin chitosan fiber membrane, with similar microstructure like the Cibotium barometz for fast blood coagulation, and efficient drug encapsulation and controlled release. Based on microfluidic technique, three-phase tubular laminar-flow that contains inner drops will be used for template synthesis of the bamboo-like ultrathin chitosan fiber membrane. Because the hemostatic effect of chitosan and the restriction of bamboo-like microstructure on the flowing of blood, the ultrathin fiber membrane enables fast blood coagulation. The bamboo-like microcavities can be used for compartmentalized encapsulation of versatile drugs for controlled release, which allows healing of the wound during the hemostasis. This work focuses on the microfluidic fabrication and the structure control of the ultrathin fiber membrane, the study of the effect of their microstructures on the blood coagulation, the investigation of their drug encapsulation and controlled release property as well as the mass transfer kinetics model, and the study of the synergetic effect and mechanism of controlled drug release and blood coagulation.
