中文摘要: MSTN基因作为肌肉生长的负调控因子,在骨骼肌生长过程中扮演着重要的角色。本研究针对国家级保护品种边鸡生长速度缓慢的缺点,利用本课题组研究发现的MSTN基因G2283A位点,建立慢长型(GG)和快长型(AA)边鸡群体。通过屠宰测定,分析MSTN基因对肌肉生长的遗传效应;制作边鸡组织切片,分析两个群体肌纤维特性的差异;采用荧光定量PCR技术,分析MSTN以及肌肉生长相关基因(MyoD、Myf5、MyoG和Smad3)在两个群体中的表达差异;培养边鸡的骨骼肌细胞,分析干扰和过表达MSTN基因对细胞增殖以及对MyoD、Myf5、MyoG及Smad3基因表达的影响。综合上述研究结果,将有助于阐明MSTN基因对边鸡肌肉生长产生遗传效应的分子机制,将鉴定的分子标记(G2283A)应用于边鸡育种建立快长型品系(AA),16周龄体重可比慢长型(GG)提高约10%,对边鸡的产业化发展具有极大的推动作用。

英文摘要: MSTN gene which functions as a negative regulator of skeletal muscle is playing an important role in the skeletal muscle growth process. In order to improve the growth speed of the national protected chicken breed Bian chicken, we will use the mutation G2283A of the MSTN gene which was found in our former research to construct fast (AA) and slow (GG) growth speed Bian chicken populations. By slaughtering test, we will analyse the genetic effect of the MSTN gene on muscle growth. Preparing tissue slice of the Bian chicken, we will compare the character of muscle fiber between the two populations. The expression differences of the MSTN and the related genes that have effects on mucle growth (MyoD, Myf5, MyoG and Smad3) will be analysed by real-time fluorescence quantitative PCR between the two populations. Cultivating Bian chicken skeletal muscle satellite cell in vitro, we will analyse the effects on skeletal muscle satellite cell proliferation, as well as the expression patterns of the MyoD, Myf5, MyoG and Smad3 genes by interfering and overexpression of the MSTN gene. Synthesizing the above research results is help to clarify the molecular mechanism about the genetic effect of the MSTN gene on skeletal muscle growth.
