中文摘要: 藏鸡是我国青藏高原特有的原始小型地方鸡种,对高海拔低氧环境具有稳定遗传的适应能力。本研究选择包括低氧诱导转录因子、细胞与生长因子以及神经递质生物合成、氧结合蛋白、血管舒张因子、葡萄糖代谢等途径中关键酶等基因作为低氧适应相关候选基因。设计探针,制备含多基因点阵的芯片,利用芯片杂交、扫描和数据标准化研究这些基因在藏鸡(以低地鸡作对照)模拟低氧孵化和常氧孵化时不同胚胎发育阶段心脏、肝脏、尿囊绒毛膜和肺脏等组织中定量表达;Bayesian Network法分析低氧适应性多基因调控网络,解析多因子之间的相互关系,分析藏鸡与低地鸡在低氧环境中胚胎发育启动和改变转录活性的基因,找到这些基因中对胚胎低氧适应调控起主导作用的一两个基因;然后进一步测定此关键基因的mRNA和蛋白表达,筛选SNP标记;揭示这些基因以及整个网络在藏鸡低氧适应中的功能,从系统生物学角度解析低氧适应的分子机制。

英文摘要: The Tibetan chicken, a unique primordial chicken breed native to Qianghai-Tibet Plateau, have steady genetic adaptation to hypoxic environment of high altitude. In present study, thirty-two genes, including hypoxia inducible factors and some key enzymes in biosynthesis of cell and growth factors, neurotransmitters, proteins carrying oxygen, vessel relaxing factors, and in metabolize of glucose, are selected as candidate genes related to hypoxic adaptation. The genes chips with multi-genes lattice are prepared with probes designed. The genes quantitive express in the different time embryonic tissues of heart, liver, chorioallantoic membrane or lung of Tibetan chicken (lowland chicken as control) incubated simulated hypoxic and normal conditions are measured with hybridization, scan and data standardization. The relationship of the thirty-two genes and the gene regulatory network are analyzed, and the genes that express promoted by hypoxia or change transcription activity in hypoxic environment are investigated to find one or two dominant genes that regulate these genes related to hypoxic adaptation. Furthermore, the mRNA and protein expressions and SNPs of the key genes are determined to reveal the function of these genes and entire network in hypoxic adaptation in Tibetan chicken, and explain ultimately the molecule mechanism in system biology.
