中文摘要: 逆转座是新基因产生的一种重要方式。mRNA被逆转录然后插入到基因组的某个位置。新插入的序列被称为逆基因。短柄草为新型温带禾本科模式植物。短柄草Bd21的全基因组测序已经完成并进行注解。本项目拟采用最新的逆基因预测方法对短柄草进行全基因组逆基因预测,分析所有短柄草逆基因的分布、结构以及功能。针对生物信息学预测的有功能的目标逆基因,进行基因的原核体外表达,确定能正确表达的目标逆基因。利用实时荧光定量PCR技术对逆基因的表达进行研究,揭示逆基因的表达规律及非生物胁迫响应模式。筛选出与逆境有关的逆基因,转化短柄草后,检测其抗逆情况。本项目将阐明短柄草逆基因的分布、结构、功能和逆基因在短柄草漫长演化过程中所处的地位等相关信息,为构建植物中逆基因演化关系图提供短柄草的重要信息,同时作为温带禾本科植物的模式植物,短柄草的逆基因信息势必为研究小麦和其他温带禾本科植物逆基因提供重要参考。

英文摘要: Retroposition is an important copy mechanism for generating new genes, named retrogene. Brachypodium is a novel model plant for structural and functional genomic studies of temperate grasses. And the line Bd21 of Brachypodium has been sequenced and annotated in 2010. In this study, all the retrogenes of Brachypodium will be obtained from the whole genome sequences by the newest method of retrogenes forecasting. The distribution, structure and function of these retrogenes will be analyzed. The prokaryotic expression of the retrogenes will be made to get the object retrogenes which can express right. The expression rules and responding model for abiotic stress will be analyzed by Real-time PCR. And the object retrogenes were transferred into Brachypodium to validate the function of abiotic stress. This project will illuminate the distribution, structure, function and the status in the evolution process of Brachypodium retrogenes. This will provide important information for constructing the evolvement relation of plant retrogenes. And as the model of temperate grasses, Brachypodium retrogenes' information will provide vital reference for the retrogene research of temperate grasses.
