中文摘要: 苜蓿根蘖性状是一种起源于黄花苜蓿的优异特性,根蘖型苜蓿具有抗寒、抗旱、耐牧的特点,是干旱半干旱草原区生态建设的优良类型。本研究针对国内外根蘖性状发生分子调控机制研究的空白,在根蘖型苜蓿新品系选育的研究基础上,采用相同遗传背景的根蘖与非根蘖苜蓿无性系为试验材料,通过根蘖性状形态发生过程中内源激素动态研究,确定取样的关键时期与组织部位,构建具有时空代表性的根蘖苜蓿转录组文库,利用RNA-seq高通量测序技术对转录组进行测序,通过差异基因表达与功能显著性富集(GO)分析,筛选出根蘖性状发生的相关基因,明确其序列特征分析与功能定位。通过本项目的实施,可阐明根蘖性状发育过程与形态建成内在机制的科学问题,探明根蘖特性与抗逆性之间的内在关系,获得根蘖性状发生相关基因的序列特征和功能定位,挖掘出优异功能基因资源,拓展根蘖苜蓿育种的技术手段。

英文摘要: Creeping-rooted trait in alfalfa was the character originate in yellow flower alfalfa which can generate shoots from the root system,and provide the plants with a rhizome-like ability to regenerate in stands.This habit of spreading by means of root proliferation provides alfalfa with a form of vegetative spread and multiplication,which has important significance to improving the ability of winter hardness,drought and grazing tolerance.To study expression of genes responsive to the creeping-rooted character,RNA-seq was used in this research to identify differentially expressed genes between the creeping-rooted plants and non-creeping-rooted plants with the same genotype. Constructing the transcriptomic library and using sequence analysis by the Illumina HiSeq? 2000,through the GO and DEG analysis,the associated genes will be sepreted .Finally,the vectors of over expression and RNAi expression were be constructed and the transferred to Arabidopsis to identify the gene function.The research will be make the molecular mechanism of creeping-rootedness of alfalfa clear, and isolating the associated gene,which provides the base for the discovering and identification of functional gene breeding of new alfalfa cultivater.
