中文摘要: 目的 研究rhEPO抑制创伤性脑水肿的分子机制。方法 体内实验:脑创伤后给于rhEPO治疗,检测脑含水量的变化,肢体运动功能,AQP4,MMP9 mRNA、蛋白的表达;使用MAPKs信号通路阻断剂,观察参与rhEPO治疗创伤性脑水肿的信号通路。体外实验:星形胶质细胞氧糖剥夺/复氧前rhEPO预处理,检测细胞水肿,AQP4,MMP9 mRNA、蛋白表达,使用MAPK信号通路阻断剂,观察参与rhEPO作用的信号通路。结果 体内实验:EPO能减轻创伤后脑含水量58%(P<0.05),提高运动功能评分(P<0.05);减轻AQP4和MMP-9 mRNA和蛋白的高表达(P<0.05);可减少ERK的磷酸化水平(P<0.05),与阻断剂阻断ERK通路减轻脑水肿的作用相同;体外实验: EPO减轻细胞体积肿胀43%(P<0.05),减轻形态学及超微结构变化,减轻AQP4和MMP-9 mRNA的高表达(P<0.05);可减少ERK,P38的磷酸化水平(P<0.05),与使用其阻断剂作用相同;结论 rhEPO通过调节MAPKs信号通路磷酸化水平及AQP4、MMP9高表达,抑制创伤性脑水肿。

英文摘要: Objective To study the molecular mechanism of inhibition of traumatic brain edema by rhEPO. Methods: After traumatic brain injury treating with rhEPO, detect changes in brain water content, motor function, AQP4, MMP9 mRNA, protein expression; using of MAPKs signaling pathway inhibitor to observe the pathway participated in rhEPO treatment of traumatic brain edema. In vitro experiments: After oxygen and glucose deprivation/reoxygenation in astrocytes pretreating rhEPO, detect cell volume, AQP4 and MMP9 mRNA, protein expression. using the MAPK signaling pathway inhibitor to observe the pathway participated in. Results: rhEPO reduce brain water content by 58% (P <0.05), improve motor function score by 48% (P <0.05), reduce high expression of mRNA and protein of AQP4 and MMP-9 (P <0.05), and reduced ERK phosphorylation levels (P <0.05), similar with blocking ERK pathway; rhEPO reduced cell volume swelling 43% (P <0.05), reduce high expression of mRNA and protein of AQP4 and MMP-9 (P <0.05),and reduce ERK, P38 phosphorylation levels (P <0.05) similar with using their antagonist. Conclusion: rhEPO inhibition of traumatic brain edema by regulating the phosphorylation of MAPKs and AQP4, MMP9 high expression.
