中文摘要: 年龄相关性黄斑变性是世界范围内的主要致盲眼病。脉络膜新生血管是该病的致盲原因,基于免疫的炎症反应在脉络膜新生血管发生过程中起到重要促进作用。IL-17是自身免疫性炎症疾病中代表性效应分子,最近的研究表明IL-17与年龄相关性黄斑变性相关,但IL-17是否在脉络膜新生血管发生过程中发挥作用及其机制尚不明确。本研究通过体外研究一方面探讨IL-17对脉络膜新生血管发生的三个关键环节-脉络膜血管内皮细胞增殖、移行和小管成形-的直接影响,并深入探讨IL-17导致脉络膜血管内皮细胞移行是否是由小分子GTP酶Mac-1介导的细胞骨架蛋白分子重排引起及其相关信号通路,另一方面探讨IL-17通过激活局部的巨噬细胞和视网膜色素上皮细胞后分泌的细胞因子对内皮细胞增殖、移行和小管成形的间接影响,以阐明IL-17如何影响脉络膜新生血管的形成及其分子机制,为药物防治脉络膜新生血管提供分子靶点。

英文摘要: Age-related macular degeneration (AMD) is a leading cause of irreversible blindness in the world. Most blindness in AMD results from invasion of the retina by choroidal neovascularization (CNV). The immune inflammation plays an important role in the development of CNV. IL-17 is a typical effector in autoimmune inflammatory diseases. Recent researches reported IL-17 is associated with AMD, but it is unclear that whether IL-17 plays a role in the initiation and the development of CNV. This study investigates the direct effect of IL-17 on the three key steps of angiogenesis-choroidal endothelial cell proliferation, migration, and tube formation. Whether IL-17-mediated choroidal endothelial cell migration results from reorganization of cytoskeleton protein mediated by Mac-1 belonged to small GTPase will be given further study. This study also investigates the indirect effect of cytokines produced by macrophage and retinal pigment epithelial cells cultured with IL-17 on choroidal endothelial cell proliferation, migration, and tube formation. The study hopefully identifies the molecular target for the medication therapy of AMD.
