中文摘要: 作为纳米科技一个引人注目的研究领域,发光量子点对无机材料化学基础研究和生物医学实际应用均有重要的意义。ZnO虽然安全无毒、价廉物美,但是通常方法制备的ZnO纳米材料发光性能都不够理想,特别是它在水溶液、细胞培养液和动物血液中的稳定性需要提高。ZnO的可见荧光不仅和它的粒径、分散性相关,更主要的是取决于纳米粒子的表面状态和内部缺陷,而这两个方面直接决定了ZnO生物荧光探针的合成制备与表面修饰的方法途径,最终影响到ZnO荧光探针穿透组织细胞、生物识别与标记、乃至药物释放的过程。最近几年,课题申请人潜心钻研ZnO纳米发光材料,已经取得了许多突破性的成果。本项目计划采用多种手段对ZnO量子点进行表面修饰和内部掺杂,制备在体外细胞环境与小动物体内稳定发光的分子探针,对常见的癌细胞进行体外识别,并且与纳米给药系统结合起来,尝试对裸鼠表皮肿瘤(皮肤癌)的诊断和治疗。

英文摘要: Photoluminescent quantum dots have received intensive attention in both basic researches of Inorganic Material Chemistry and practical applications of Biomedical Science. ZnO is a kind of safe, nontoxic and cheap material, but the photoluminescent properties of the conventional ZnO nanoparticles are not satisfying, especially in water, cell culture and animal blood. ZnO luminescent properties not only depend on the particle size and dispersion, but also rely on particle surface state and internal defect. The latter two aspects determine the routes of ZnO syntheses and modifications, and finally influence the processes of biological labeling and drug delivery of ZnO fluorescent probes. In the past years, the project applicant devoted himself to studying ZnO luminescent nanomaterials and achieved progress in this field. In the present project, the applicant plans to modify ZnO externally and dope ZnO internally, and then prepare stable fluorescent probes in vivo and in vitro for distinguishing cancer cells and deliver drugs to skin tumor of nude mice.
