中文摘要: 过度的内质网应激可以导致软骨细胞的凋亡,通过TRAF-2 和GADD153启动细胞凋亡基因,引起钙离子通道活化,Ca2+离子平衡紊乱,进一步激活下游的caspase级联反应,促进软骨细胞凋亡。本课题前期实验证实透骨消痛胶囊能够抑制软骨细胞的凋亡,从而延缓关节软骨的退变,但是是否通过内质网应激而达到抑制其凋亡的作用,是本课题研究的重点。因此本课题采用体外实验观察透骨消痛胶囊对内质网应激下关节软骨细胞存活状态、功能变化、GADD153、BiP、ATF-4及caspase效应组的影响,从多个层面探讨其减轻内质网应激,抑制软骨细胞凋亡的作用机制和作用靶点,为临床应用透骨消痛胶囊治疗骨性关节炎提供理论依据。

英文摘要: Excessive endoplasmic reticulum stress can lead to the apoptosis of chondrocytes,it activates apoptotic genes by TRAF-2 and GADD153, caused by calcium channel activation of Ca2+ balance disorders, further activation of the caspase cascade downstream to promote chondrocytes apoptosis. The subject's early experiments confirmed that Tougu Xiaotong capsules can inhibit the apoptosis of chondrocytes, thereby delaying the degeneration of articular cartilage, but whether to inhibit apoptosis through the endoplasmic reticulum stress, is the focus of this research project. Then this subject in vitro experiments to observe Tougu Xiaotong capsules endoplasmic reticulum stress, articular cartilage cell survival status, functional changes, GADD153, of BiP, ATF-4 and caspase-effect group influence, and explore its mitigate the multiple dimensions of endoplasmic reticulum stress, inhibition of chondrocyte apoptosis mechanism and the role of targets, and provide a theoretical basis for clinical application of Tougu Xiaotong capsules treatment of osteoarthritis.
