中文摘要: 胰腺癌是一种高度恶性的消化系统肿瘤,目前尚无有效的化疗药物能够明显改善患者的预后。我们在前期研究中发现,超营养剂量的亚硒酸钠能够明显抑制胰腺肿瘤细胞的生长,诱导细胞产生凋亡和自吞噬。此外,我们还发现Wnt/β-catenin通路中的关键分子及其下游靶基因同样发生了显著的变化。这些发现提示我们Wnt/β-catenin通路在硒诱导胰腺癌发生程序性死亡及其转换中起到了重要的调节作用。本研究将致力于验证该假说并阐明其中的具体作用机制。我们拟利用LSL-KRASG12D;Pdx1-Cre转基因小鼠模型和SCID小鼠肿瘤模型在体内环境下验证亚硒酸钠对胰腺肿瘤的作用,为进一步将硒应用于胰腺肿瘤的预防和治疗提供理论基础和实验依据。

英文摘要: Pancreatic cancer is a highly aggressive tumor of digestive system and there is no effective drug can obviously improve the patient's prognosis. Our preliminary studies find sodium selenite of super-nutritional doses could inhibit the growth and induce the apoptosis and autophagy of pancreatic cancer cells. In addition, the key molecular and target genes of Wnt/β-catenin signaling have changed markedly. All these findings prompt that Wnt/β-catenin pathway play an important role in selenite-induced programmed cell death and their transformation. Our research committed to verify the hypothesis and clarify the specific mechanism. We intend to use LSL-KRASG12D;Pdx1-Cre transgenic mice and SCID mice model to test the function of sodium selenite to pancreatic cancer. All these researches will provide us the theoretical basis and experimental evidence in the application of selenite with prevention and treatment of pancreatic cancer.
