中文摘要: 藏民居住区多处高海拔,盛行其间的藏医在脑缺血防治方面功效独到,尤以二十五味珊瑚丸疗效最佳,前期大鼠急性脑缺血实验均已证实,但组方成分复杂。本项目拟在前期建立的三种骨髓间充质干细胞模型上,运用交叉追踪探寻方中对脑缺血确有疗效的20个有效成分,并筛选出效果最好的3个,重点研究3个活性物质对神经元保护、抑制细胞凋亡和炎症反应等信号通路的影响,以揭示其神经元保护和/或再生作用的机制;再从调节脑组织突触可塑性和细胞间化学信息传递的角度研究这些功效物质的神经修复本质及作用靶点。本项目的研究结果有望建立一种完整研究体系、获得高效的单体化合物并阐明其作用机理;将从行为药理学、细胞生物学和分子神经生物学等层次推动藏医脑缺血名方的研究发掘,并有望直接发现具有神经保护或再生作用的新药源头分子。

英文摘要: Most of Tibetans inhabit in high-altitude areas , the Tibetan medicine have effects on preventing and treating cerebral ischemia,especially Er Shi Wu Wei San Hu Wan . The early experiments of rats with acute cerebral ischemia have confirmed positive results in Tibetan medicine , however , the components are complex .In this project , with the use of three kinds of bone marrow mesenchymal stem cell model, we want to find 20 effective components on the curative effect of cerebral ischemia by using the method of cross track, and to screen for the best 3 effective components. Our study will focus on the signaling pathways effects of three active substances on protecting neurons, inhibiting cell apoptosis and inflammatory reactions. In the end we will reveal its neuroprotective and / or regenerative mechanism.To study these efficacy substances on the nature of nerve repair and targets from chemical information transfer regulation of brain synaptic plasticity and cell angles. The results of this project is expected to establish a complete research system, obtain monomer compound efficiently and to clarify the mechanism of cerebral ischemia .Our work will promote the research of exploring the Tibetan medicine on the fields of the pharmacology, cell biology , molecular neurobiology , and the project helps to find the molecular as a direct source of new drug which has neuroprotective or regenerative effect.
