中文摘要: 研究发现,在氮饥饿条件下,硅藻细胞PSⅡ复合体的光合效率显著降低,这预示着硅藻PSⅡ复合体的结构及其在类囊体膜上的空间排布发生了变化,从而影响了它的生理功能。本研究以硅藻金色奥杜藻和三角褐指藻为材料,采用条件可控的光生物反应器培养并获取正常和氮饥饿条件下的藻细胞,制备PSⅡ复合体,利用双向电泳及质谱分析技术比较其蛋白组成差异;采用多种电泳技术、HPLC和圆二色光谱技术研究PSⅡ复合体的亚基结构及色素组成;通过稳态光谱分析PSⅡ复合体的光谱特性及氮对PSⅡ复合体能量传递的作用规律;并借助扫描电镜探究PSⅡ复合体在类囊体膜上的排布特征。通过比较氮饥饿和正常培养条件下硅藻PSⅡ复合体结构与功能的差异,分析氮饥饿对硅藻光合效率影响的作用机理,在PSⅡ复合体水平上揭示硅藻光系统结构与功能对氮素营养的响应机制。该研究对阐明氮影响光合作用效率的分子机理具有重要意义。

英文摘要: It was illustructed that the photosynthetic efficiency of PS Ⅱ complex in diatom was obviously decreased under nitrogen starvation condition. This phenomenon indicated the change of the structure and arrangement of PSⅡ in thylakoid which could effect its function. This research use Odontella aurita and Phaeodactylum tricornutum as material and culture with manageable photoreactor, the algae under normal and nitrogen starvation conditons are collected. PSⅡ complex is isolated and the protein constitution is compared using two-dimensional electrophoresis and mass spectrometry technology. Meanwhile, the multiple electrophoresis technology, HPLC and circular dichroism spectrum is used to investigate the subunit structure and pigment composition. Furthermore, steady spectrum is used to analysis the spectrum characteristic of PSⅡ complex and the energy transfer mechanism of nitrogen. Finally, the PSⅡ complex arrangement is studied using the scanning electron microscope. By comparing the structure and function difference of PSⅡ complex under control and nitrogen starvation conditions, the response meanchanism of how nitrogen starvation effect the photosynthetic efficiency will be illustrated. This study is important for clarifying the mechanism of nitrogen effect on photosynthesis efficiency.
