中文摘要: 磷是土壤中植物生长发育的主要限制因子之一。植物在进化过程中形成了一套适应低磷环境的机制。作为在转录水平的基因表达调控因子,转录因子发挥着关键作用。申请人前期工作中筛选到了水稻 WRKY 家族缺磷响应转录因子基因,将其命名为 OsWRKY1,初步结果显示该基因可能通过调控下游磷吸收转运相关基因以维持水稻的磷素动态平衡。此外,我们发现该基因亦显著影响水稻的生长发育过程。本项目拟利用已经获得的OsWRKY1过量表达和突变体植株,同时采用生物信息学分析、基因芯片、ChIP和EMSA等手段筛选出受OsWRKY1直接调控的下游基因,明确OsWRKY1在维持水稻磷素动态平衡过程中的功能及其作用机制。本研究有望阐明水稻中首个参与维持磷素动态平衡的WRKY家族转录因子的生理功能及其分子机制,为培育磷高效作物提供候选基因。

英文摘要: Phoshporus (P) is often a limiting factor in soil for plant growth and development. Plants have evolved a suite of adaptive strategies to cope with low phosphate (Pi) conditions. It is transcription factors that play crucial roles in such transcriptional regulation. In my previous work, a Pi-starvation responsive transcription factor from the WRKY family was identified in rice, and designated as OsWRKY1. The physiological role of OsWRKY1 in maintaining rice Pi homeostasis was investigated. In addition, OsWRKY1 was found to affect rice growth and development. In the present work, we intend to study the physiological roles of OsWRKY1 in maintaining P homeiostasis in rice by using OsWRKY1 overexpressing and knock-down or knock-out plants. Simultaneously, in an attempt to dissect the regulatory mechanisms of OsWRKY1, we shall also utilize bioinformatic analysis, microarray, ChIP (Chromatin Immuno-Precipitation) and EMSA (Electrophoretical Mobility Shift Assay) to screen the downstream target gene directly regulated by OsWRKY1. We believe we will stand a good chance to elucidate the important role and underlying molecular mechanism of the first WRKY transcription factor involved in maintaining P homeostasis in rice, providing candidate gene for molecular breeding of high P-use-efficiency crops.
