中文摘要: 杆状病毒是一种很有应用前景的微生物杀虫剂,其包埋型病毒(ODV)负责昆虫的口服感染,但ODV囊膜的形成机制尚不清楚,其中内核膜如何出芽形成核内微囊泡(ODV囊膜的前体)更是未见相关报道。我们的前期研究发现:分别缺失杆状病毒保守基因p48、ac93和ac76均导致病毒在感染细胞中不能形成微囊泡,装配成熟的病毒核衣壳也不能出核。我们推测这3个基因的缺失影响了核纤层的局部解聚,阻碍核膜流动性的改变。核纤层通过可逆性解聚调节有丝分裂过程核膜的崩解和重建;在疱疹病毒感染的细胞中核衣壳出核也需要核纤层的局部解聚。本项目拟通过分子生物学、病毒学和免疫组织化学等技术和手段,研究杆状病毒感染是否诱导核纤层局部解聚,p48、ac93和ac76与核纤层解聚的关系以及作用方式,分析微囊泡的形成机制,为揭示杆状病毒的复制、装配机制和流行病的发生规律,更好地利用杆状病毒进行害虫生物防治提供重要的理论基础。

英文摘要: Baculoviruses are applied as promising microbial pesticides. The occlusion-derived viruses (ODV) of baculovirus are responsible for primary recognition and invasion in the insect gut. So far, the mechanism for the morphogenesis of ODV envelope remained unclear. The processes that the inner nuclear membrane buds into the nucleoplasm to form intranuclear microvesicles, which are the precursors of ODV envelopes, have not been elucidated. Our previous studies have demonstrated that, in p48-, ac93- or ac76-knockout bacmid-transfected cells, neither intranuclear microvesicles nor nucleocapsids egressing from nuclei were observed. During mitosis, the reversible depolymerization of nuclear lamina contributes to the breaking and reconstruction of the nuclear envelope; moreover, partial disassembly of lamina is required for the nuclear egress of capsids in herpesvirus-infected cells. Thus, we hypothesize that P48, Ac93 and Ac76 may alter the fluidity of nuclear membranes by regulating the partial disassembly of lamina. In this project, we plan to investigate whether lamina undergoes partial disassembly upon baculovirus infection and the role of p48、ac93 and ac76 in this process.
