中文摘要: 活体单细胞免标记信息检测与获取是目前细胞科学追求的目标,细胞本征信息检测正在成为实现这一目标的关键,这其中尤以电/力学等本征信息的检测与获取为当前的研究热点。然而,由于缺乏高通量自动检测获取方法,制约着电/力学本征信息作为生物标记的使用。针对上述问题,本项目拟开展基于光电耦合场作用的活体单细胞电/力学本征信息高通量自动获取方法研究。运用光电耦合场方法,探索光诱导电液动力学机制下的活体单细胞多参量作用响应与调控理论;开展基于光诱导介电泳机理的活体单细胞操控理论研究,建立光诱导介电泳场致作用下的活体单细胞电/力学本征信息获取模型;研究活体单细胞显微视觉动态观测与分析方法,提出具有统计学特性的大样本量活体单细胞电/力学本征信息高通量自动获取量化方法。通过本研究,将发展一种可高通量自动获取大样本量活体单细胞本征信息的系统方法,该方法对推动生物物理和细胞科学研究具有特别重要的意义。

英文摘要: Currently, label-free information detection and acquisition of living individual cells is one of the main aims of cell research community, among which intrinsic cell information detection especially the electrical and mechanical information of the cells has become the key to achieve this target. However, the lack of high-throughput and automatic detection approaches restricts the application of using this kind of information as biomarkers. To address this issue, we here propose a high-throughput and automatic approach based on the theoretical study of living individual cells using the optical-electric coupling field. Briefly, the multi-parametric response and control of living individual cells will be explored, followed by the theoretical study of manipulating living individual cells using the optically-induced dielectrophoresis. Based on these results, modeling methods leading to the intrinsic cell information will be established accordingly. Furthermore, dynamic observation and analytic methods of microscopic vision for living individual cells will be investigated, and a high-throughput and automatic approach with statistical characteristics of large population cells will be described. In summary, in this proposal, we will develop a systematic method for the automatic acquisition of intrinsic information of living individual cells in large populations with the merit of high-throughput. The proposed cellular information acquisition approach will have special significance for the development of biophysics and cell research.
