中文摘要: 中药药性理论是中药核心理论,是指导中医临床用药的重要组成依据,利用现代科技手段对传统中药药性理论进行科学阐述已成为中药理论研究重要课题之一。前期大量研究表明中药药性的“寒”、“热”与机体的能量代谢具有密切的关系。不过,以往的研究多集中在某一代谢相关的酶、代谢终产物等下游产物及动物行为表现,从转录组水平分析药性调节机体能量代谢的研究却不深入。然而,转录过程恰恰是机体对药性最直接的反应,而且能量代谢涉及的生理生化过程很多,从组学角度出发能够全面考察寒热药性对机体能量代谢的影响,深入分析各能量代谢途径对药性的应答及相互之间的作用关系。本研究以典型寒热中药处理的大鼠模型为研究对象,从转录组水平考察中药寒热药性对大鼠能量代谢的影响,并筛选中药寒热药性相关生物标志物。同时,建立基于细胞培养的、以筛选得到的生物标志物表达变化为检测指标的中药寒热药性评价模型,为揭示中药寒热药性作用机理提供理论与实践基础。

英文摘要: The theory of Chinese herbs nature is the basis and guidance for traditional Chinese medicine, so it is one of the most important problem that how to elaborate the traditional theory by using modern technology. There were lots of evidence showed that the “hot” (or “cold”) and the energy metabolism were closely related. While, previous studies on the energy metabolism regulated by the Chinese herbs natures mainly paid attention to certain enzyme, metabolites and the behaviors of experimental animal, and rarely involved in transcriptome. But transcription is the direct response for the effect of drugs, and transcriptome proivded comprehensive analysis about the change of energy metabolism effected by the herbs nature and the relationship between the herbs nature and the pathway of energy metabolism. Based on transcriptome, this project will study the change of energy metabolism of rat effected by the cold and hot nature of tranditional Chinese medicine, screen the potential biomarker for cold or hot nature, and build an cell-culture-based herbs nature evaluation model. The research of this project will provid experimental evidences and theoretical basis to reveal the mechanism of nature of traditional Chinese medicine.
