中文摘要: 白斑综合症病毒(white spot syndrome virus, WSSV)长期困扰着世界对虾养殖业,虽然对该病毒已开展了广泛的研究,但目前仍未发现其侵染的关键途径。c-Jun氨基末端激酶(c-jun N-terminal kinase, JNK)信号通路是信号传递网络中与免疫密切相关的途径之一,在基因表达和细胞凋亡调控中发挥着重要作用。本研究组利用多种细胞信号通路激酶抑制剂进行筛选比较,首次发现JNK特异性抑制剂对抑制WSSV增殖效果最为明显,并通过体内基因沉默实验进一步证实了对虾JNK信号通路与WSSV的侵染存在着密切的关系。因此JNK细胞信号通路很可能成为了解WSSV感染机制的切入点。本项目将对对虾JNK细胞信号通路展开深入的研究,探讨JNK 细胞信号通路在对虾白斑综合症病毒感染中的生物学功能及相关分子机理,寻找阻断WSSV感染的潜在靶点,从而为对虾WSSV病毒的防治提供新途径。

英文摘要: White spot syndrome virus(WSSV) is one of the most devastating and it can cause massive death in cultured shrimp. Although extensive research has been carried out, but its key mean of infection has not been found. c-Jun N-terminal kinase(JNK) is a serine/threonine kinase which is critical for signal transduction pathways. It has been implicated in regulation of gene expression and apoptosis. Our lab through the screening of kinase inhibitors first discovered JNK inhibitor significantly inhibited the proliferation of WSSV and had most significant effect. Gene silencing experiments in vivo have verified the results. This demonstrated that JNK signaling pathway played an important role during WSSV infection. Therefore, this project will further elucidate molecular mechanism of JNK signaling pathway in WSSV infection and provide new ideas and effective way for possible disease prevention and control.
