中文摘要: 草鱼是我国产量一直居首的重要养殖鱼类,但迄今仍未选育出良种,性成熟周期长是制约草鱼良种选育亟需解决的关键问题之一。鱼类生殖细胞移植技术(即鱼类“借腹生子”技术)是细胞工程育种的一个分支,该技术是将供体鱼的外源生殖细胞移植到宿主鱼的腹腔中,利用宿主鱼繁育供体鱼的后代。本项目拟以性成熟周期长的草鱼作为供体,以性成熟周期短的近缘赤眼鳟等作为宿主,采用免疫组化、显微注射移植、分子标记等方法研究宿主仔鱼体内的原始生殖细胞的发生,转移状态,生殖细胞移植的最佳时期及影响生殖细胞移植技术的关键因子(移植细胞的剂量、移植后生殖细胞的嵌合率及影响移植的关键因子)等,筛选出具有草鱼生殖细胞的阳性宿主,获得生殖细胞移植的最佳参数,同时可建立一个鱼类生殖细胞移植技术研究平台。在该平台下通过种内,种间生殖细胞的移植,探索生殖细胞移植技术在缩短草鱼性成熟周期的可行性。

英文摘要: Germ cell transplantation (refer to surrogate broodstock) as one brench of cell engineering breeding is one of the most hot international topic, however no more domestic report concerning it existed. This technology is to transplant germ cells from donor fish to recipient fish by intraperitoneal injection of new hatched larves. It can be used in shortening generation time, conservation of valuable and/or endangered species and monosex male breeding in fish. In this proposal we will use grass carp as donor while Squaliobarbus curriculus and hybrids between Ctenopharyngodon idellus and Squaliobarbus curriculus as recipient to do the transplantation of germ cells. We will study the position and transfer profile of primordial germ cells and some key factors in the process of the transplantation. The establishment of transplantation system in freshwater teleosts would allow the creation of a new broodstock system in which a target species with long generation time could be produced from related species with short generation time。We will study the establishment of a working model for interspecies and interspecies germ cell transplantation (GCT) in freshwater fish.
