中文摘要: 医药行业对人肝实质细胞的需求已经成为一个亟待解决的问题。建立人源化肝脏的猪模型有利于扩增人肝实质细胞和由干细胞诱导产生的人肝细胞,为再生医学和药物筛选提供手段。本课题将首次建立人源化肝脏的猪模型。通过应用非整合型慢病毒结合锌指核酸酶技术,在西藏小型猪的胚胎成纤维细胞中敲除延胡索酸乙酰乙酸脱氢酶(Fah)基因。应用核移植克隆技术获得Fah(-/-)纯合子猪。应用基因、蛋白和生理性状等方法鉴定Fah(-/-)猪的预期表型并繁育扩增。Fah(-/-)猪将被用于同种和人的肝细胞移植研究,以期获得猪和人的肝细胞再殖,最终获得人源化肝脏的猪模型。并利用改进的荧光素酶标记供体肝细胞技术实现无创伤追踪监测供体细胞在受体内的归巢和增殖行为。为提高人肝细胞的再殖,将应用特殊的免疫抑制剂降低人肝细胞在受体猪中的免疫排斥,实现人肝功能在猪肝中的重建。由此制备的人源化肝脏和扩增的人肝细胞可应用于医药研究和器官移植。

英文摘要: Hunam hepatocytes and large animal models with humanized livers,such as pig,are more and more required in today's research and apllication in biomedical field. The goal of this application is to achieve a pig model with humanized liver by using several novel techniques. Non-integrated lentiviral vector combining with zinc finger nuclease technique will be used to target fumarylacetoacetate hydrolase (Fah) in fetal fibroblasts of Tibet Minipig for generating Fah knockout Fah (-/-) fibroblasts. The nuclei of Fah (-/-)fibroblasts will be used to generate Fah(-/-) pig by using somatic nuclei tranfer technique. The Fah(-/-) piglets are expected to be obtained finally and will be proved by the results of deletion of Fah in genmoic DNA, losing Fah protein expression and losing Fah relative metabolic activities. The clony of Fah (-/-) pig will be expended for following researches. Liver repopulation in Fah(-/-) pig will be proved in transplantation assay by using singenic pig hepatocytess as donor cells. The Fah (-/-) pigs will be treated with relavant immune suppression agents for achieving immunodeficiency for xeno-engraftment of donor human hepatocytes.Donor hepatocytes will be labeled by luciferase for non-invasive detection of liver repopulation with luminescent imaging system.
