中文摘要: 利用巨磁阻抗(GMI)效应传感器和磁性标签研究面向心血管疾病生物标志物肌红蛋白(Mb)、C-反应蛋白(CRP)的新型快速检测方法。采用MEMS技术制备带材和多层膜GMI传感器;从电动力学和铁磁学出发建立带材和多层膜GMI效应的分析模型,对影响GMI传感器性能的因素如形状、结构、尺寸等进行研究;采用MEMS技术制备微流控部件;采用MEMS技术设计、制造可控的Au单元及其分布;研究多种自组装膜技术,通过在传感器表面上的自组装膜固定心血管疾病生物标志物单克隆抗体,采用双抗夹心法实现磁珠在传感器上的连接;研究磁场作用下磁珠磁化产生的磁场对GMI传感器性能的影响;通过对磁珠的浓度、分布等的研究,获得GMI传感器探测心血管疾病生物标志物的灵敏度和探测极限等信息,最终获得心血管疾病生物标志物的检测方法,为研制新型重大疾病早期诊断和预警系统提供科学依据。

英文摘要: New rapid detection method of biomarkers of cardiovascular disease based on giant magneto-impedance sensor and magnetic labels is investigated.GMI sensor based on soft magnetic ribbons and magnetic thin films is fabricated by MEMS technique.Analytical model of GMI effect is established from electrodynamics and ferromagnetics,the effects of sensor shape,structure,size and so on on the GMI effects are studied sysmatically. Microfludic component, and controllable Au element and its distribution on sensor surface are fabricated by MEMS technique. Technique of self-assembled monolayers is investigated, biomarkers of cardiovascular disease such as C-reactive protein (CPR),Myoglobin (Mb) antibody is immbolized on the self-assembled monolayer of the sensor surface using the Au elements,the labeled magnetic beads and antigen are immobilized on the sensor surface by using the immunoassay reaction. Magnetic field produced by magnetic beads under the external magnetic field on the performance of the GMI sensor is studied. By studying the concentrations,numbers and distribution of magnetic beads on performance of the GMI sensor, sensitivity and detection limit of biomarkers such as CRP, Mb antigen are obtained,which providing a novel detection method for diagnosis and alert system of cardiovascular disease.
