中文摘要: 组蛋白修饰在植物生长发育和抗逆等生命过程中起着重要作用。近年来植物应答低温胁迫的分子机制的研究已经取得了很大进展,但基于组蛋白修饰的调控机制在冷驯化中的研究还知之甚少。前期工作中,我们发现了拟南芥RCC1家族基因TCF1(Tolerant to Chilling and Freezing1)受低温诱导表达,其无义突变体tcf1比野生型更加耐冷;TCF1蛋白与核心组蛋白结合,通过组蛋白修饰调控下游基因表达,发挥其在冷信号通路中的作用。但是TCF1基因自身在冷驯化中的表达调控模式与组蛋白修饰之间的调控机制尚不清楚。已有数据表明TCF1基因的表达受组蛋白H3K27me3甲基化水平调控,本研究将详细探讨组蛋白H3K27me3甲基化修饰,甲基化修饰酶以及TCF1基因上游调控蛋白ICE1与TCF1基因表达之间的调控关系。本研究将对深入理解组蛋白修饰参与调控植物的冷信号通路有一定的理论意义。

英文摘要: Histone modification plays important roles in plant development and plant coping with abiotic stress. Recently, some molecular mechanism progresses on low temperature in plants have achieved. While the roles of histone modification in plants response to cold stress remain largely unknown. We had studied the function of an RCC1 (Regulators of Chromosome Condensation1) family like protein TCF1 (Tolerant to Chilling and Freezing1) which null mutation tcf1 mutant showed a cold tolerance phenotype compared with wild type.TCF1 interacts with core histones and affects histone modification of its downregulated genes. We focus on the relationship between TCF1 gene expression and histone modification. TCF1 gene is a target of H3K37me3, and TCF1 gene expression regulated by low occupation of histone H3K37me3. Further study will exam whether histone modification, methyltransferase/demethyltransferase and ICE1 protein involve in regulating TCF1 gene expression. Our research will provide some evidences for histone modification in plant response to cold stress.
