中文摘要: 心衰是严重威胁着人类的健康和生命。心衰的发生是一个复杂的病理性过程,多种因素参与其中,不仅取决于DNA序列,环境、饮食和生活习惯等外在因素同样可以作用于生物。高脂食物的摄入是目前导致心血管病发病人数持续增加的一个重要诱因。已知饮食可以改变生物的表观遗传修饰,而表观遗传学修饰与心衰的发生发展密切相关。因此,研究心衰发生的表观遗传调控是刻不容缓的工作。果蝇遗传学工具丰富,便于定时定点特异性敲低或者敲除基因的表达,是研究心衰分子机制的理想模型。本项目已初步确定高脂食物诱发果蝇心衰发生,心脏特异性敲低表观遗传调控基因HDAC3导致相似的心衰表型。在此基础上,我们利用果蝇成体心脏功能分析系统,研究高脂食物是否通过作用于表观遗传调控子HDAC3诱导心衰发生以及它们的分子机制,帮助人类认识饮食对心衰的影响,为临床上有效预防和治疗心力衰竭奠定新的理论依据。

英文摘要: Heart failure is a serious threat to health in face of mankind. Heart failure is a complicated pathological process involving many factors. The susceptibility to heart failure depends on not only DNA sequence, but also your diet, lifestyle and the environment you exposed to by causing epigenetic changes. High-fat diet is currently main reason of increasing incidence of heart failure. Thus, investigating epigenetic regulation of heart failure is urgent. And epigenetic is closely related to heart failure. The Drosophila is the excellent model to study molecular mechanism of heart failure, owing to the easy manipulation of genetic tool. Our previous study found that high-fat diet cause heart failure, and cardiac specific knock down of HDAC3 give rise to the similar heart phenotype. Taking advantage of drosophila adult heart research platform, we would further study the hypothesis that high-Fat Diet inducing heart failure via epigenetic regulation of HDAC3 and their molecular mechanism, to help people realize the dietary effects on heart failure and provide theoretical basis for clinical prevention and therapy of heart failure.
