中文摘要: T-2毒素是A类单端抱霉烯族真菌毒素中毒性最强的一种,对人类和动物的健康造成了严重危害,至今未找到有效的解毒与治疗方法。本课题以小鼠睾丸支持细胞(TM3)为研究对象,深入研究T-2毒素对TM3细胞的损伤作用,重点分析氧化应激介导的线粒体途径在T-2毒素诱导TM3细胞凋亡过程中的作用。在此基础上利用基因沉默和基因克隆技术构建表达不同水平CyPD蛋白的TM3细胞模型,CyPD是一种线粒体伴侣蛋白,能够调控线粒体通透性转换孔(MPTP)。T-2毒素染毒后,评价不同水平的CyPD蛋白对细胞线粒体功能、线粒体通路、死亡受体通路、内质网通路及细胞凋亡的影响,深入探讨CyPD蛋白及线粒体通路在T-2毒素生殖毒性发生和发展过程中的作用,阐明T-2毒素产生毒性作用的生物学基础,从而为防治由霉菌毒素引发的畜禽繁殖障碍提供新思路和新手段。

英文摘要: Among the type A trichothecenes, T-2 toxin has the greatest cytotoxicity, may pose a threat to human and animal health around the world. Until now, there is no effective detoxification and treatment measures for T-2 toxin poisoning. To investigate the potential mechanism of Cyclophilin-D and mitochondrial pathway in T-2 toxin poisoning, TM3 cells will be a model to evaluate the toxic effects of T-2 toxin, focus on oxidative stress-mediated mitochondrial pathway in T-2 toxin induces apoptosis. Then, gene silencing and gene cloning technology is being use to build the model of the TM3 cell expression different levels CyPD, which is a mitochondrial chaperone that has been shown to regulate the mitochondrial permeability transition pore (MPTP). TM3 cells were treated with T-2 toxin, to explore the significance of?level of CyPD effects mitochondrial function, mitochondrial pathway, death receptor pathway, endoplasmic reticulum pathway and apoptosis. The research results will provide new ideas and new means for the prevention and treatment of livestock reproductive failure caused by mycotoxins.
