中文摘要: 细胞DNA损伤应激反应是一个很复杂的过程,最终可导致细胞周期停滞、DNA修复或凋亡,但其中的机制还未完全明确。我们在前期研究中发现在DNA损伤的细胞中有剪接因子SR家族基因的高表达,并伴随变异剪接活性的增高,还证明CD44和Fas分子发生了变异剪接。但是对于在此过程中是否有其他基因发生变异剪接,以及发生变异剪接的基因在DNA损伤应激反应中的作用还不清楚。因此我们拟利用全基因组变异剪接分析技术,系统地鉴定在DNA损伤中发生变异剪接的基因,并对鉴定的部分基因在DNA损伤后对细胞周期、DNA修复以及凋亡的影响进行分析,从而阐明变异剪接在DNA损伤应激中的作用

英文摘要: Cellular DNA damage response (DDR) is a very complex process, which eventually can lead to cell cycle arrest, DNA repair, or apoptosis. However, the underlying mechanism is still not fully clear. Previously we have found that in DNA-damaged cells the expression of splicing factor SR family members was increased, as well as the splicing activity. We further showed that alternative splicing occurred for CD44 and Fas. Still, whether alternative splicing occurs to other genes during this process, or the funtion of the alternativel spliced genes remain unknown. Therefore, we intend to use the whole-genome analysis technology to systemetically identify those alternative spliced genes during DDR, and evaluate the effects of some identified genes on cell cycle, DNA repair and apoptosis, in an effort to elucidate the function of alternative splicing in DDR.
