中文摘要: 本课题利用微流控芯片电泳/激光诱导荧光或激光共聚焦扫描显微镜等技术和镉系量子点(Cd-QDs)自身荧光以及镉离子与荧光探针作用后产生的特征荧光,建立单细胞中Cd-QDs、镉离子原位测定的新方法,研究Cd-QDs在细胞内镉离子的释放规律;探究细胞内Cd-QDs、镉离子的剂量-效应关系与作用机理;比较相同条件下Cd-QDs与镉离子毒性效应的大小、异同点并阐明原因。进一步探明Cd-QDs和镉离子诱发细胞氧化应激、FAS或MAPK相关信号通路变化在细胞凋亡中的作用与机理。为与镉系污染物相关的疾病早期诊断及高危人群预警提供技术支持,为不同暴露途经下研究结果的评判提供参考依据。本研究成果主要以论文的形式发表,预计发表10篇以上高水平科研论文,其中影响因子大于4的4-6篇;申请专利2-3项;培养博士研究生3-4名,硕士研究生8-10名。

英文摘要: By utilizing micro fluidic chip electrophoresis coupled with laser-induced fluorescence or laser scanning confocal microscope et.al techniques and the autofluorescence of Cd-QDs and the characteristic fluorescence derivated by cadmium ion, this research planned to build up a new strategy to quantify the contents of Cd-QDs and cadmium ion in situ respectively. The release rule of cadmium ion from Cd-QDs within single cell will be proved. This research aims to explore the dose-response relationship and mechanism in cells. We will try to compare and explain the differences and similarities of the toxicity between Cd-QDs and cadmium under the same condition. Furthermore, this research intended to explore the toxicity and mechanism of oxidative stress, MAPK signaling pathway changes in cell apoptosis induced by Cd-QDs and cadmium. This research may provide techniqucal support for early diagnosis of disease and high risk population and references to judge the results obtained from different exposure approaches. To solve the above mentioned scientific questions, this research are expected to apply 2-3 invention patents, publish more than 10 SCI research papers (impact factors of 4-6 papers will be greater than 4), culture 8-10 masters and 3-4 Ph D students.
