中文摘要: 杜梨原产我国,是优良的耐非生物胁迫砧木。前期的工作以耐逆的杜梨株系DL-518为材料,利用高通量测序技术分析发现盐胁迫24h后,HD-Zip转录因子显著富集且转录水平显著上调。在此基础上,我们拟以DL-518为材料,通过PCR扩增法克隆PbHBs基因,并分析不同胁迫下PbHBs基因的时空表达模式,筛选与渗透胁迫相关性最大的PbHBs,以此为目标基因,利用转基因和病毒介导的基因沉默技术获得过表达和基因沉默烟草,从而鉴定PbHBs功能,同时利用荧光定量RT-PCR技术分析渗透胁迫杜梨及转基因植株时,ABA途径关键基因NCED、PP2C、PYR/PYL、SnRK2以及下游功能基因OSM、PR2、PR3、PR4、GLU的表达模式,从而揭示HD-Zip转录因子调控杜梨耐渗透胁迫的分子机理,为梨及其它木本植物育种提供理论研究和实际应用价值的基因资源。

英文摘要: Birch-leaf pear (Pyrus betulaefolia), a pear species originating from China. In our previous works,transcriptome analysis by high-throughput sequence using DL-518,a pear strain with high torelant to high salt and drought stress,revealed significant richment and up-regulated expression of HD-Zip transcription factors after 24h with 150 mM NaCl treatment.The induced expression of PbHB by high salt and drought stress was confirm by Real-time PCR and semi-quantitative RT-PCR.Based on this work,RT-PCR amplification will be used to clone PbHBs sequences from DL-518.The expression patterns of PbHBs in different time points and different tissues under high salt and drought stress treatment will be monitored.PbHBs,which display significant positive correlation with osmotic stress, will be transformed into tobacco by transgenic technology and virus-induced gene silencing (VIGS).Overexpression transgenic plants and gene silencing plants with PbHBs will be as material to evaluate the function and analysize the regulating mechanism of PbHBs on adaptation response of plants to osmotic stress by monitoring expresion patterns of NCED,PP2C,PYR/PYL,SnRK2 and OSM, PR2,PR3,PR4 and GLU gene.This applies not only to agricultural crops, but also to woody plants that are gaining importance as renewable resources。
