中文摘要: 在高等真核生物中,染色质绝缘子参与基因的时空表达需要通过与其相结合的蛋白质CTCF来实现。目前为止,我们对染色质绝缘功能的调控机制仍不清楚。我们和其他科学家发现染色质的绝缘功能不仅依赖于CTCF本身,还依赖于与其相互作用的部分蛋白质,如:RNA解螺旋酶p68和粘结蛋白复合物。为了鉴定出更多参与调控染色质绝缘功能的蛋白质,本项目在已有研究工作的基础上,通过蛋白亲和纯化及互作分析,发现浓缩素II型复合物存在于CTCF的蛋白复合物中。本研究将对浓缩素II型蛋白复合物的生物学功能展开研究,重点阐明浓缩素II型蛋白复合物如何调控CTCF介导的染色质绝缘功能和远距离染色质相互作用,并对其调控的内在分子机理进行深入探索。该研究有助于我们更深入的理解CTCF及其蛋白复合物对染色质绝缘功能的调控机制,为染色质三维空间结构对基因的表达调控提供新的理论依据。

英文摘要: In higher eukaryotic organisms,the chromatin insulator is involved in gene spatial and temporal expression through its combination with insulator binding protein CTCF. So far, the regulatory mechanism of these factors on chromatin insulation is still unclear. We and other scientists discovered that chromatin insulation not only depends on CTCF itself, but also depends on the factors which interact with CTCF (For example, RNA helicase p68 and Cohesin complex). In order to identify novel proteins to participate in CTCF-mediated chromatin insulation, based on our recent research work, we identified that Condensin II complex exists in CTCF protein complexes through protein affinity purification and protein-protein interaction analysis. In this study, we will study the biological functions of Condensin II complex, focusing on how condensin II complex regulates CTCF-mediated chromatin insulation and long range chromatin interaction, and the intrinsic molecular mechanisms of regulation. This study will help us to deeply understand regulatory methanisms of chromatin insulation mediated by CTCF and its interacting partners, to provide a new theoretical basis for the three-dimensional structure of chromatin on the regulation of gene expression.
