中文摘要: 神经胶质细胞对神经元的发育、功能与健康至关重要,然而胶质细胞对神经突触传递、突触可塑性、感觉生成与行为有什么实时的影响目前仍不明确,在体研究结果尤为缺乏。我们拟联合采用光遗传学、在体膜片钳电生理记录、在体胞内钙成像检测,结合分子遗传学、行为学分析、正反向遗传学筛选等方法,基于秀丽线虫(C.elegans)在体研究体系,探讨以下问题:1)激活神经元对AMsh和CEPsh胶质细胞的实时影响及其分子机制;2)激活和抑制AMsh和CEPsh胶质细胞对神经元活性、突触传递、感觉生成和行为的影响,及其分子细胞和环路机理。本课题将从新的视角探讨胶质递质的生理意义及其作用机理,揭示神经元- - 神经胶质细胞相互作用的核心分子机制,阐述神经胶质细胞在感觉生成和行为调控中的作用,为人类神经胶质细胞和神经功能的研究奠定基础,进而为慢性痛、癫痫、抑郁症、神经退行性疾病等相关神经精神疾病的诊断与治疗创造理论条件。

英文摘要: Glial cells actively participate in neuronal development, function and health. However the function of glial cells in synaptic transmission, synaptic plasticity, sensory formation and behavior generation remain poorly understood. Particularly,studying neuron-glia interaction in vivo are extremely challenging. Caenorhabditis elegans (C.elegans) is an ideal model organism for in vivo studies with fast genetics. We plan to employ combined approaches of molecular genetics,optogenetics,electrophysiology,calcium imaging and behavioral analysis, to address: 1) The effects of neuronal activity on the AMsh and CEPsh glial cells; 2) The effects of AMsh and CEPsh glial cell activity on neuronal activity, synaptic transmission, sensory generation and behaviors. We also seek to identify key molecules involved in above processes and the underling mechanisms. This project will better define the roles of C.elegans glial cells in sensory generation and behaviors. It also provides a fantastic opportunity to indentify the core molecular mechanism of neuron-glia interaction in higher organisms and has the potenial to provide new insights into possible diagnostic tools and treatments for chronic pain, epilepsy, depression and other neuropsychiatric disorders.
