中文摘要: 乳腺癌相关基因的突变在乳腺癌发生发展中起着重要的作用。多项研究证明蛋白酪氨酸磷酸酶与肿瘤的生物学行为改变密切相关。我们前期研究采用二代深度测序技术检测原发及转移性乳腺肿瘤组织,结果提示新发现的蛋白酪氨酸磷酸酶受体D基因(PTPRD)突变与三阴性乳腺癌的转移相关,但具体作用机制还有待进一步探讨。基于前期研究成果,我们推测PTPRD编码的磷酸酶因外显子突变引起其功能异常以及与下游蛋白异常酪氨酸磷酸化而促进乳腺癌转移。我们将通过从分子、细胞、组织以及动物水平等多方面探讨该基因突变在乳腺癌转移中的作用及可能机制,探索JAK2/STAT3和PI3K/AKT靶向抑制剂在含有PTPRD突变的三阴性乳腺癌中的潜在价值。本研究从磷酸酶基因突变这个角度为揭示三阴性乳腺癌转移机制提供支持,以突变基因及其相关通路作为乳腺癌转移的标记物及干预靶点,为三阴性乳腺癌患者个体化治疗提供依据。

英文摘要: Numerous evidence indicates that breast cancer-associated gene mutation play an important role in breast cancer initiation and progression. Based on previous research, protein tyrosine phosphatase have relationship with breast cancer metastasis and poor prognosis. Our subsequent next generation deep sequencing of the full length PTPRD revealed novel PTPRD gene mutations in prime and metastatic triple negative breast cancer (TNBC), which may play important role in breast cancer cell migration. However, PTPRD gene mutation promote cell migration and the mechanism is still unclear. The mutation of protein tyrosine phosphatase may lead to abnormal phosphorylation of downstream molecules. Then this property correlates with the solid cancer metastasis. To demonstrate our hypothesis, we use various biochemical and biological assays from different levels of molecular, cells, tissue and animal. We also plan to evaluate the inhibitor of JAK2/STAT3 and PI3K/AKT in TNBC with PTPRD mutation. In conclusion, gene mutation and related signaling pathways are opening avenues of research in triple negative breast cancer metastasis, which are potential markers and intervention targets. The research may provide the basis for individualized treatment of triple negative breast cancer patients.
