中文摘要: 为进一步提高肺癌(尤其是非小细胞肺癌)治疗靶向性和增强抑瘤效应,本项目在以往肿瘤基因放射治疗研究的基础上,拟采用辐射激活效率高的Egr-1启动子,诱导野生型p53和内皮抑素(endostatin)基因表达,发挥其促肿瘤细胞凋亡及抗肿瘤血管生成作用,并通过针对表皮生长因子受体(EGFR)的双组分靶向性腺病毒载体,显著提高肿瘤细胞感染率,联合放射治疗发挥靶向性抑瘤作用。通过观察辐射诱导基因表达的量效和时效规律、融合蛋白的分泌表达特点及其联合放射治疗对体外人非小细胞肺癌细胞株和裸鼠体内移植人非小细胞肺癌荷瘤模型的抑制作用,制定基因联合辐射治疗肿瘤的最佳方案,并进一步探讨其抗肿瘤作用机制。本研究成果将建立一种更为有效的肺癌综合治疗模式,为肿瘤基因-放射治疗向临床应用转化过渡提供理论基础和实验依据。

英文摘要: In order to enhance the specific anti-tumor effect in the protocol of non small cell lung cancer, the construction of recombinant adenovirus vectors Ad.Egr-wtp53-endostatin and Ad.CMV-sCAR-EGF will be carried out. The expression of wild type p53 and endostatin genes could show the most significant inhibition of tumor growth and angiogenesis induced by the radiation-sensitive promoter Egr-1 in non small cell lung cancer cells. The infection of non small cell lung cancer cells could be mediated with the bicomponent targeted adenovirus vector to aim directly at epidermal growth factor receptor (EGFR). The targeted anti-tumor effect will be induced by the gene-radiotherapy in non small cell lung cancer cells. In this research, the time- and dose-effects of radiation-induced genes expression will be analyzed. The therapeutic effect of gene therapy combined with radiotherapy will be observed in vivo and vitro. The mechanism of anti-tumor effect in non small cell lung cancer gene-radiotherapy will be investigated and the optimized therapeutic regimen of non small cell lung cancer will be determined. This research may set the stage for developing rational clinical protocols in non small cell lung cancer treatment.
