中文摘要: 黄瓜植株性型分化丰富,是研究植物性别决定的模式植物。以M基因的克隆为代表的单性/两性花调控机理研究刚刚起步,是有待进一步深入探索的问题。前期项目组收集了两个新型黄瓜两性花突变体XN38和SHZ-H。本项目拟利用图位克隆技术分离黄瓜材料XN38和SHZ-H中两性花控制基因m-38和m-shz,并分析其生化功能和基因表达模式;配制并筛选双突变个体,以及与m基因结合的三重突变体,分析基因互作关系;将M基因、M-38基因和M-SHZ基因转化拟南芥,尝试构建黄瓜单性/两性花基因拟南芥检测体系;进行转基因拟南芥和转基因黄瓜材料性型分化特异时期RNA-Seq差异分析,分析其中基因调控路径。最后结合所有已克隆的单性/两性花控制基因,以及测序获得的相关基因,构建黄瓜单性/两性花基因调控网络模型。本研究旨在探索黄瓜雄蕊发育受抑制的分子机制,将黄瓜单性/两性花调控机理研究推向深入,丰富黄瓜性型分化研究。

英文摘要: As a result of its diversity in sex types, cucumber (Cucumis sativus L.) has been used as a model for sex determination in plants. Although the cloning of M gene opens a window for the regulation in unisexual/bisexual flowers, the detail mechanism is still a problem needed to be further explored. Our research team has collected two new bisexual cucumber mutates, XN38 and SHZ-H. In this study, with the map-based cloning strategy, we plan to (1) isolate the genes (m-38 and m-shz, respectively) controlling the bisexual flower phenotypes in the XN38 and SHZ-H lines, then analyze their biochemical function and gene expression pattern; (2) develop and screen the double and triple mutation to study the genes interaction; (3) transform the M gene, M-38 gene and M-SHZ gene into Arabidopsis thaliana, and try to construct a detection system of cucumber unisexual/bisexual genes in Arabidopsis plants; (4) do RNA-Seq analysis with transgenic Arabidopsis and cucumber plants to isolate the related genes in the regulation of unisexual/bisexual flowers in cucumber. Finally, with the three unisexual/bisexual flowers controlling genes, as well as the related genes from RNA-Seq, we will attempt to construct the gene network model for the regulation of unisexual/bisexual flowers in cucumber. The purpose of this project is to study the arrest phenotype of the stamen develpoment in cucumber plant, make some further studies in unisexual/bisexual flower regulation, and enrich the research of sex determination in cucumber plant.
