中文摘要: 基因治疗是维持骨缺损局部生长因子有效治疗浓度进而提高疗效最有希望的一种方法,但基因工程在骨缺损的治疗中尚存在以下不足:种子细胞来源受限制、单一目的基因缺乏协同性、病毒载体存在免疫原性和安全性等很多问题。本研究采用高性能壳聚糖纳米微囊包裹非病毒载体多基因转移技术,通过设计壳聚糖纳米微囊-双基因(BMP-2/bFGF)复合体,转染于ADSCs(Adipose Derived Stem Cells,脂肪源性基质细胞,脂肪干细胞),研究其成骨性能。本研究是骨缺损基因治疗的技术改良,将为基因工程人工骨的制备提供新思路和理论依据,并最终为人工骨移植修复骨缺损提供新途径。

英文摘要: Gene therapy is the most promising way to maintain effective concentration of growth factor and thus improver the efficacy of treatment to local bone defects. But there are still some deficiencies when gene engineer goes to application on the treatment of bone defects. For example, limited source of seed cells, lack of coordination of single gene, immunogenicity and safety of viral vectors. In our study, the capacity of bone formation will be evaluatied after the co-transfecion in ADSCs(Adipose Derived Stem Cells) through the combination of chitosan nanocapsules and double gene(BMP-2/bFGF). This research is a improved technology for genetic therapy of bone defects and can provide new methods for prepation of artificial bone through genetic engineering, ultimately, it may find new way for artificial bone graft to repair bone defects.
