中文摘要: miRNA通过对靶标基因表达的调控,在癌症等疾病和生命过程中发挥重要作用。miRNA靶标的预测是miRNA研究的瓶颈和热点问题。针对miRNA靶标预测算法过度依赖序列信息而且性能不高的现状,本项目从基因调控角度入手,提出"miRNA→作用通路→转录因子→调控基因集"因果假设模型,并利用系统生物学思想开展靶基因预测算法的研究。本项目拟采用统计分析、机器学习、贝叶斯推断、基因富集度分析等方法,有机地融合序列信息、多种生物学实验数据、作用通路、转录因子调控信息以及其它生物学先验知识,在获得更高预测准确度的同时,对预测结果做出合理生物学解释,为后续的鉴定工作提供有力参考。本项目将为miRNA靶标预测提供新的思路,对阐明miRNA作用机制、揭示miRNA功能有重要意义。

英文摘要: miRNAs play important regulatory roles in many diseases such as cancer and biological processes by regulate the expression of their target genes. Targets prediction is the bottle-neck of miRNA researches. Considering the situation that most target prediction algorithm excessively rely on the sequence information and result in very poor performance, this project will work on systems biological approach, which based on a novel causality hypothesis which is "Pathway→Transcription factor→regulation gene set", to contribute to miRNA target prediction. This project aims to integrate sequence level information, variety of biological experiments data, Pathway information, Transfactor information and other biological prior knowledge. Hopefully, this research will provide strong reference to the following miRNA target identification by improving the performance target prediction algorithm as well as assign proper biological explanation to the prediction results. Therefore, this research will draw a new light to the miRNA target prediction, and means a lot to discover the mechanism as well as the function of miRNA.
