中文摘要: 核小体是真核染色质的基本重复单位,核小体核心组蛋白的N末端尾部可以发生多种组蛋白修饰。"组蛋白密码"提出多个修饰组合在一起共同发挥生物学作用,相关的研究结果显示一些组蛋白修饰之间存在组合关系并且影响基因表达。但是,能够形成组合关系的修饰之间的因果关系仍然没有明确,修饰之间因果关联的研究对于理解组蛋白修饰组合意义重大。为了通过分析修饰之间的因果关系研究组蛋白修饰组合,我们首先通过实验弥补现有酵母组蛋白修饰数据库的不足。在建立相对完善的酵母组蛋白修饰数据库的基础上,构建稳定的修饰贝叶斯网络。借助网络明确组蛋白修饰之间的因果关联,进而通过修饰间的因果关联探索和分析组蛋白修饰组合。同时,通过实验验证部分组蛋白修饰因果组合,为在分子水平解释修饰组合提供更加可信的证据。我们的研究拟在分子生物学层次揭示相关组蛋白修饰酶之间的作用关系,有助于阐明组蛋白密码的产生。

英文摘要: Nucleosomes are the fundamental repeating units of eukaryotic chromatin. Various kinds of histone modifications occur on the N-terminal tails of core histones of nucleosomes. The 'histone code' suggests that multiple histone modifications act in a combinatorial fashion to play biological function. The relevant research results show that there are combinatorial relationships among some histone modifications and the relationships affect gene expression. But the causal relationships among the modifications for which there are combinatorial relationships are not be confirmed yet. The study on the causal relationships among the modifications is of great significance to gain insight into the combinations of histone modifications. In order to study the combinations of histone modifications by analyzing the causal relationships among the modifications, we firstly make up for the shortage of existing histone modification database. The stable Bayesian networks of the modifications will be constructed based on relatively consummate database of histone modifications established in yeast. The network can illustrate the causal relationships among the modifications. Then the combinations of histone modifications will be explored and analyzed on the basis of the causal relationships.
