中文摘要: Rops(植物Rho GTP酶)是植物体内最重要的信号传导分子开关之一,参与调控植物正常生长、发育以及植物对环境胁迫的响应。我们已有的研究结果表明,在玉米植株中瞬时沉默玉米Rop1基因(ZmRop1)可促进甘蔗花叶病毒(SCMV)的系统侵染,在原生质体中瞬时超量表达ZmRop1则抑制SCMV基因组复制,推测ZmRop1调控了玉米抗SCMV的防卫反应。本项目拟在前期工作的基础上,利用反向遗传学和分子生物学手段,寻找并鉴定与ZmRop1互作的寄主因子,深入研究ZmRop1及其互作因子对SCMV复制、移动的影响,揭示ZmRop1调控玉米抗SCMV的分子机制,为最终采用基于调控措施创制具有持久SCMV抗性的玉米新种质材料提供理论依据。

英文摘要: The Rop (Rho-related GTPases from plants) family belongs to the Ras superfamily of small GTPases. Like other small GTPases, Rops are versatile plant signalling regulators involved in pathways that influence growth and development and the adaptation of plants to various environmental situations. Our previously study indicated that transiently silencing ZmRop1 facilitated SCMV systemic infection in maize plants, transient over-expression of ZmRop1 in maize protoplasts reduced SCMV viral accumulation; speculating that ZmRop1 involves in regulating maize resistance to SCMV infection. In this project, we will use reversed genetics and molecular biology tools to elucidate the influence of ZmRop1 and its interactors on the SCMV viral multiplication, movement and their roles in the signaling pathways of maize defence responses signaling pathway to SCMV infection, to further elucidate the mechanism by which ZmRop1 mediates maize resistance to SCMV. This project will serve for establishing theoretical basis to create maize germplasm materials with durable SCMV resistance.
