中文摘要: 肉是人类蛋白质等营养素的主要来源,摄入不同的肉类蛋白质可导致肠道微生物的变化,也会引起机体代谢的变化,但肉类蛋白质摄入-小肠吸收-大肠肠道微生态变化-肝脏代谢之间内在联系仍不清楚。本项目采用大鼠模型,分别饲喂猪、牛、鸡和鱼肉蛋白质90天后,取动物肝脏、血液、小肠黏膜、肠道食糜和粪便,分析小肠食糜中蛋白质消化产物的组成及肠黏膜pept1变化,并应用宏基因组学技术分析肠道微生物区系的变化,结合大肠食糜中短链脂肪酸的测定和微生物体外培养实验,探究不同肉蛋白质摄入对大鼠肠道微生物区系变化的内在机制;应用转录组学(包括芯片技术)和蛋白质组学分析肝脏相关蛋白质(尤其是脂质代谢通路相关调控因子)的基因表达,结合生理生化指标(如胰岛素、甲状腺素、脂蛋白等)的测定,探究不同肉蛋白质摄入对肝脏代谢影响的分子机制。研究结果将为机体健康的膳食调控提供一定的科学依据。

英文摘要: Meat is a very important source for human nutrients, especially of proteins. The ingestion of meat proteins from different species has been reported to be associated with changes in intestinal microbiota and metabolism. However, the interactions among dietary proteins, intestinal absorption, intestinal microbiota and liver metabolism are still unclear. This project is designed to profile intestinal microbiota and their metabolites after the ingestion of proteins derived from pork, beef, chicken and fish. Liver, blood, intestinal contents and faeces will be sampled after 90 d feeding. In-vivo digesta of proteins in small intestine will be analyzed using LC-MS-MS. Microorganisms and short-chain fatty acids in the colon content will be profiled using meta-genomics and GC-MS. Association of dietary proteins and their digesta with intestinal microbiota could be realized on the basis of in-vitro cultivation of intestinal microorganisms. Metabolic regulatory factors in liver (for example regulatory factors related to lipid metabolism) will also be analyzed using transcriptomics and proteomics. The relation of dietary proteins to molecular regulation of lipid metabolism in liver will be realized on meta-analysis of insulin, thyroxin, lipoprtoeins. The results are expected to be helpful for health maintenance via dietary regulation.
